IDENTIFICATION OF BIOMARKERS IN CRUDE OIL OF 
KAZAKHSTAN USING
GAS CHROMATOGRAPHY – MASS SPECTROMETRY

Crude oil is a non-renewable source of energy. The importance of crude oil 
have become the main reason to determine the origin of petroleum, which 
describes its physical properties and chemical composition, and this knowledge 
helps to determine the quality of crude oil 
Biomarkers are one of the most important hydrocarbon groups in crude oil 
used for chemical fingerprinting. They are complex molecules that have been 
derived from former living organisms. Biomarkers can be used to fingerprint 
spilled oils because they have most of the original carbon skeleton which helps to 
identify the conditions for crude oil’s origin. The origin of crude oil is considered 
to the main scientific problem nowadays. Crude oil fingerprinting technology is the 
main method for oil spill identification. In comparison with other hydrocarbons, 
biomarkers are very resistant to degradation. As all crude oils have their own 
specific origin under the wide variety of conditions, they may have a unique 
biomarker fingerprint. Therefore, it is extremely important to analyze biomarkers 
which may have significant information about crude oil’s origin peculiarities. Gas 
chromatography is considered to be the most effective method for biomarkers 
analysis because it provides good separation of complex compounds while mass-
spectrometry detector allows to identify these compounds within the sample. 
The aim of this research work was to identify biomarkers that crude oils 
contain. In accordance with the aim, crude oils from Western and Southern 
Kazakhstan were analyzed. Before the analysis they were fractionated using 
column chromatography into two fractions: saturated and aromatic. Analysis was 
provided by gas chromatography with mass-spectrometry: 
During this research work biomarkers such as hopanes, bisnorhopanes, 
isocopalanes, 
pregnane, 
androstane, 
androstane, 
(5β)-allopregnane, 
D-
homopregnane, (5α)-cholestane, stigmastane were found in crude oils. For all 
crude oils Pr/Ph and Pr/C17 ratios, tricyclic/pentacyclicterpanes and hopane ratio, 
sterane distribution were calculated. According to Pr/C17 and Ph/C18 ratios almost 
all West Kazakhstan crude oils are resistant to biodegradation.
